Comparison of two analytical methods (electrophoresis and HPLC) to detect thalassemias and hemoglobinopathies

چکیده
The role of the laboratory in the diagnosis of thalassemia and hemoglobinopathies is crucial. The objective of our study was to compare two common methods used in hemoglobinopathy and thalassemia investigation. Hemoglobin electrophoresis and HPLC (high performance liquid chromatography) were used to investigate patients suspected with thalassemia. A total of 301 adult and child blood samples were examined for routine thalassemia screening with both methods. The HPLC method and electrophoresis methods gave similar results for patients without thalassemia. HbH disease was readily noted on HPLC but was difficult to detect on electrophoresis. There were some changes in classification for patients with possible β thalassemia minor. For patients with β thalassemia the HPLC gave better results for low HbA2 concentrations.

Thalassemias and hemoglobinopathies are highly prevalent in Thailand and other Southeast Asian countries. Accurate and precise separation of hemoglobin types, together with reliable quantitation, are essential for differential diagnosis of these diseases. Presented in this study is a multicenter validation of a fully automated capillary electrophoresis (CE) method for hemoglobin separation and ...

Hemoglobinopathies are most common inherited disorders in the world; approximately 7 percent of the worldwide population and 5-6 percent of population of Iran are carriers. The hemoglobin disorders inherit as autosomal recessive and are very common in the Mediterranean area and much of the Asia and Africa. The control of this inherited disorders need to genetic counseling and accurate screening...
متن کاملHigh-performance liquid chromatography in the diagnosis of hemoglobinopathies and thalassemias. Report of three cases.
High-performance liquid chromatography is a technic that has recently been applied to the diagnosis of hemoglobinopathies and thalassemias. Its advantages over other methods include increased sensitivity, resolution and simplicity, as well as speed. In this report, the authors present an HPLC procedure that uses a weak cation exchange column and a gradient elution system for the diagnosis of he...
